چکیده

The ethylene glycol bis(P-aminoethyl ether)-N, N’-tetraacetic acid-sensitizing factor extracted from natural actomyosin at low ionic strength was concentrated in the fraction obtained between 40 and 60% ammonium sulfate saturation. Fractionation within narrower salt limits and at low protein concentrations yielded a troponin-rich fraction at 40 to 53% saturation and a tropomyosin-rich fraction at 53 to 60% saturation. These ammonium sulfate fractions could be further purified by isoelectric precipitation at pH 4.6. The supernatant of the 40 to 53 % fraction was shown to contain primarily troponin, while the precipitate of the 53 to 60% fraction was pure tropomyosin. It was found that for maximal EGTAsensitizing factor activity a ratio of tropomyosin to troponin of 1: 1.3, w/w, was required. This combination appears to form a complex which sediments as a single peak in the ultracentrifuge with a sedimentation constant of 5.15 S.

Involvement of envelope-bound calcium in the transient depolarization of the Escherichia coli cytoplasmic membrane induced by bacteriophage T4 and T5 adsorption.

We previously showed that adsorption of bacteriophages T4 and T5 to their respective outer membrane receptors induced a partial depolarization of the cytoplasmic membrane. As these membrane potential changes were independent of phage properties, we proposed that phage adsorption triggered the emission of a signal which must be transmitted between the two membranes. We show here that these two p...

Requirements for extraction of polyribosomes from plant callus cultures.

A procedure was developed to isolate polyribosomes from plant cell cultures. Tobacco callus extracted in 10 mm MgCl(2), 80 mm KCl, 250 mm sucrose, and 140 mm tris-HCl (pH 8.2) yielded larger amounts of polysomes than cells extracted in higher or lower ionic strength or pH buffers. Optimal conditions for extraction of polysomes from soybean callus were identical except the most suitable pH for r...

Ca(II)-calmodulin regulation of fungal dimorphism in Ceratocystis ulmi.

We have shown that Ca(II) ions, ethylene glycol-bis(beta-aminoethyl ether)-N,N-tetraacetic acid, LaCl3, and six known calmodulin inhibitors shift the yeast-mycelium dimorphic potential of Ceratocystis ulmi. Our data are consistent with the conclusions that Ca(II)-calmodulin interaction is necessary for mycelial growth in C. ulmi and that the absence of this interaction leads to the yeast phase.

Activation of the alternative complement pathway by Naegleria fowleri.

Naegleria fowleri amoebae were lysed by adult fresh human serum, and their multiplication was inhibited in culture medium supplemented with 10% fresh human serum. Heat inactivation (56 degrees C, 30 min) of serum abrogated these lytic and inhibitory effects. Absorption of human serum with amoebae failed to reduce immunoglobulin levels, and no specific antibody was detected in untreated or treat...

New calcium indicators and buffers with high selectivity against magnesium and protons: design, synthesis, and properties of prototype structures.

A new family of high-affinity buffers and optical indicators for Ca2+ is rationally designed and synthesized. The parent compound is 1,2-bis(o-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid (BAPTA), a relative of the well-known chelator EGTA [ethylene glycol bis(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid] in which methylene links between oxygen and nitrogen are replaced by benzene rings....
